SPEAKER_01We're talking about NAD, short for nicotinamide adenine denucleotide. It sounds complex, but Alex, why is everyone suddenly talking about this molecule? And what does it actually do in our bodies?
SPEAKER_00Great question, Jordan. NNAD plus is absolutely fundamental. Think of it as a vital coenzyme present in every single cell. It's crucial for two main things: turning the food we eat into cellular energy and repairing damaged DNA. Without enough NAD, our cells simply can't function effectively.
SPEAKER_01So it's like the essential fuel for our cellular engines and the repair crew for our genetic blueprint. But if it's so important, why are we talking about supplementing it?
SPEAKER_00That's where the aging connection comes in. As we get older, our systemic NAD plus levels naturally decline. By middle age, many of us have only about half the NAD plus we had in our youth. This drop isn't just a side effect of aging. Scientists now believe it's a primary driver of many age-related dysfunctions.
SPEAKER_01So, less NAD plus means less energy, less DNA repair, and potentially faster aging? That definitely sounds like something worth addressing. And that's where these precursors like NR and NMN come into play, right?
SPEAKER_00Exactly. NR or nicotinamide ribicide and NMN, nicotinamide mononucleotide are compounds that our bodies can use as building blocks to create more NAD plus. They're designed to replenish those dwindling cellular coffers.
SPEAKER_01So if I just take an NR or NMN supplement, my NAD plus levels go up, and suddenly I'm feeling younger and repairing DNA? Is it that straightforward?
SPEAKER_00Not quite as simple as suddenly younger, Jordan, but yes, studies consistently show that oral supplementation with both NR and NMN is generally safe and effectively raises NAD plus levels in human blood. This is a critical first step because it proves they survive digestion and get into our system.
SPEAKER_01Okay, so the supplements work at increasing NAD plus in the blood. But what about the actual benefits? Are there human studies showing real impacts on health and aging?
SPEAKER_00That's where the nuances come in. While animal studies showed spectacular results, human benefits are often more subtle. Research suggests the most pronounced effects are seen in what we call replete populations. Individuals who are already experiencing some metabolic stress or age-related decline.
SPEAKER_01So, for an older person or someone with, say, pre-diabetes, it might make a noticeable difference. But for a healthy 20-year-old athlete, maybe not so much?
SPEAKER_00Precisely. For younger, perfectly healthy individuals, the returns might be diminishing. In older adults, however, studies have noted potential improvements in muscle insulin sensitivity and a reduction in systemic inflammatory markers. Think of it more as a restorative intervention than a performance enhancer for the already optimized. That makes sense.
SPEAKER_01It's about bringing things back to a healthy baseline. But our cells don't just make NAD. How do these precursors actually get converted into the active form our body uses?
SPEAKER_00Our bodies have a clever salvage pathway. Instead of building NAD plus from scratch every time, our cells efficiently recycle it. NR enters the cell and is converted into NMN, which is then converted into NAD. NMN can also enter certain cells directly through specialized transporters.
SPEAKER_01So essentially NR and NMN are just different entry points into the same NAD plus production line. And once we have more NAD, what does it actually do?
SPEAKER_00Once formed, NAD Plus acts as crucial fuel for proteins like sertuins, often called the guardians of the genome. Sertuins regulate gene expression and help our cells resist stress. NAD Plus also powers PARP enzymes, which are responsible for repairing damaged DNA. So it's both a protector and a repair man?
SPEAKER_01It sounds like increasing the supply of NAD plus is only half the battle. Is there anything else that might be leaking our NAD plus supply?
SPEAKER_00You've hit on a crucial point, Jordan. One of the biggest shifts in recent NAD plus research is the focus on an enzyme called CD38. It's a major consumer of NAD. As we age and experience chronic inflammation, CD38 levels tend to rise, essentially burning through our NAD plus supply before it can be used for repair energy.
SPEAKER_01So we're not just trying to fill the bucket, but also plug the holes. Are there strategies to inhibit CD38?
SPEAKER_00Exactly. This has led to a dual track strategy. Researchers are exploring compounds like apogenin, found in parsley and chamomile, for their ability to suppress CD38 activity. So the idea is to combine a precursor to increase supply with a CD38 inhibitor to decrease waste.
SPEAKER_01That's a much more sophisticated approach than just taking more of a supplement. What about other biochemical interactions? Does boosting NAD plus play nice with everything else in our body?
SPEAKER_00It's important to consider the methylation cycle. When our body processes NAD please and its precursors, it produces a byproduct that needs to be methylated to be excreted. This process consumes methyl groups, which are vital for many other bodily functions.
SPEAKER_01So, high doses of NAD plus precursors could potentially deplete our body's methyl group reserves? That sounds like an imbalance we'd want to avoid.
SPEAKER_00Yes, some researchers suggest co-administering a methyl donor, like trimethylglycine or TMG with high dose NAD plus precursors. This helps ensure those pathways remain balanced. It's a good example of how interconnected our biochemistry is.
SPEAKER_01Beyond supplements, are there any natural ways we can boost our NAD plus levels, or are we completely reliant on these compounds?
SPEAKER_00Absolutely not. In fact, our body has built-in mechanisms that predate modern supplements. Exercise is one of the most potent natural NAD plus boosters. Physical activity increases the enzyme NAMPT, which drives the NAD plus recycling pathway, making our bodies more efficient.
SPEAKER_01So, regular movement is still foundational. What about diet or eating patterns?
SPEAKER_00Time-restricted feeding and calorie restriction have also been shown to influence NAD plus levels. When the body senses fewer nutrients, it shifts into a state of cellular maintenance and repair, a process largely governed by increased NAD plus and sertuin activation. These lifestyle interventions are crucial foundational elements.
SPEAKER_01So, the most effective approach is likely a combination of these lifestyle factors, possibly with targeted molecular support. That makes a lot of sense. How do people even know if these things are working for them?
SPEAKER_00That's a challenge. Most available blood tests measure NAD plus in red blood cells, which is a proxy but may not reflect levels in critical organs like the brain or heart. NAD plus levels also fluctuate throughout the day. It's still an area of active research.
SPEAKER_01So a lot of it comes down to subjective metrics, then, like energy levels, cognitive clarity, and exercise recovery, alongside the evolving clinical data, of course.
SPEAKER_00Exactly. We're in an era of guided self-experimentation for many biohackers. But the key takeaway is this both NR and NMN are effective at raising systemic NAD plus levels in humans, and the human data for metabolic health and inflammatory response is increasingly promising.
SPEAKER_01So it's not a magic bullet, but a valuable tool for specific populations, especially as part of a broader, comprehensive lifestyle strategy.
